The method is characterized by incorporating a nucleotide analog having fluorescent and phosphorescent groups into the 3' terminus of a growth-initiating nucleic acid chain during the amplification of a nucleic acid molecule duplex to be tested to determine the nucleotides at the 3' terminus of the nucleic acid molecule to be tested by detecting fluorescence and phosphorescence signals.
